PrimerArray series for disease pathways

When performing gene expression analysis by real-time PCR, ready-to use primer sets for RT-PCR save time, minimize pipetting, and reduce error. Included in the PrimerArray series are sets of real-time RT-PCR primers for analyzing the expression of genes associated with specific disease states in human (Cat.# PH011–PH015) or mouse (Cat.# PN011–PN015). These PrimerArray RT-PCR primer sets allow efficient workflows for colorectal cancer gene expression analysis as well as other diseases such as pancreatic cancer, prostate cancer, melanoma, small lung cell cancer, non-small lung cell cancer, asthma, rheumatoid arthritis, and Type I or Type II diabetes.

Each array contains 96 primer pairs representing 88 genes associated with a disease pathway and 8 housekeeping genes. When comparing an unknown sample to a control sample, gene expression differences can be measured using the relative quantification method. The expression levels of multiple genes can be screened simultaneously.

The PrimerArray Analysis Tool* (download) and/or Multiplate RQ (for use with the Thermal Cycler Dice Real Time Systems, not available in all geographic locations) are useful for analyzing data obtained using these products. This analysis tool is linked to the KEGG (Kyoto Encyclopedia of Genes and Genomes) database, allowing results to be visualized on a pathway map. Primer set information and a manual on how to use the tool can be downloaded by opening the product table and clicking on the Resources tab for your product of interest.

Procedural overview for gene expression analysis by real-time RT-PCR

The overall workflow for the PrimerArray sets is straightforward:

  1. RNA extraction (30 min): We recommend NucleoSpin RNA kits (Cat. #740955.50) or RNAiso (Cat.# 9108).
  2. (Products not available in all geographic locations.)
  3. Reverse transcription (20 min): We recommend PrimeScript RT Master Mix (Perfect Real Time) (Cat.# RR036A).
  4. Dispense real-time PCR reaction solution (10 min): Prepare a master mix of your cDNA sample and TB Green Premix Ex Taq II (Tli RNase Plus) (Cat.# RR820A) and dispense into a 96-well plate.
  5. Add PrimerArray primers (5 min): Use 4 µl per reaction.
  6. Real-time PCR (1–2 hours): Compatible with all commonly used real-time PCR systems.
  7. Data analysis (30 min): PrimerArray Analysis Tool makes data analysis easy.
