Gene expression analysis to screen for pluripotency of embryonic stem (ES) cells and induced pluripotent stem (iPS) cells is streamlined and efficient with PrimerArray embryonic stem cell arrays. These arrays include primers for key markers of mouse or human ES cell pluripotency as determined by the International Stem Cell Initiative International (ISCI). When performing gene expression analysis by real-time RT-PCR, these ready-to-use primer sets save time, minimize pipetting, and reduce error.

Each PrimerArray embryonic stem cell array contains 96 primer pairs representing 88 genes associated with embryonic stem cell or iPS cell pluripotency and eight housekeeping genes. The PrimerArray Analysis Tool for Embryonic Stem Cells (available for download here) and/or Multiplate RQ (for use with the Thermal Cycler Dice Real Time Systems, not available in all geographic locations) are useful for analyzing data obtained using these products.

Procedural overview for stem cell gene expression analysis by real-time RT-PCR

The overall workflow for the PrimerArray sets is straightforward:

  1. RNA extraction (30 min): we recommend NucleoSpin RNA II kits (Cat. # 740955.50) or RNAiso (Cat. # 9108, not available in all geographic locations)
  2. Reverse transcription (20 min): we recommend PrimeScript RT Master Mix (Perfect Real Time) (Cat. # RR036A)
  3. Dispense real-time PCR reaction solution (10 min): prepare a master mix of your cDNA sample and TB Green Premix Ex Taq II (Tli RNase Plus) (Cat. # RR820A) and dispense into a 96-well plate
  4. Add PrimerArray primers (5 min): use 4 µl per reaction
  5. Real-time PCR (1–2 hours): compatible with all commonly used real-time PCR systems
  6. Data analysis (30 min): PrimerArray analysis tool makes data analysis easy
