Principle / Procedure
NucleoSpin® 8 / 96 Blood is designed for the parallel isolation of genomic DNA from whole blood. After lysis of blood cells genomic DNA is selectively bound to a silica membrane while contaminants are washed away effectively. Elution is possible even under vacuum without cross-contamination. Due to its plate design and the possibility to perform the complete procedure under vacuum NucleoSpin® 8 / 96 Blood can be easily adapted to common liquid handling instruments for fully automated processing without manual interactions.
Required hardware
Vacuum processing The NucleoSpin® 8 / 96 Blood kits can be used manually with the NucleoVac 96 vacuum manifold. For vacuum processing of the 8-well Strips the reusable Starter Set A, containing Column Holders A and NucleoSpin® Dummy Strips is required. For automation on laboratory platforms with standard 96-well plate vacuum chambers the use of the Starter Set A is also required.
Purification of genomic DNA from whole blood with NucleoSpin® 8 Blood Genomic DNA has been purified from human blood (200 μL each, frozen for six weeks at -80 °C) using NucleoSpin® 8 Blood manually under vacuum. 18 μL out of 200 μL eluate have been analyzed on a 0.7 % TAE agarose gel. M: λHindIII
Purification of genomic DNA from animal blood with NucleoSpin® 96 Blood Genomic DNA was extracted manually from pig blood (50 μL, stabilized either with EDTA or citrate) using NucleoSpin® 96 Blood with an average yield of about 1.60 μg. 20 μL of the eluate were analyzed on a 0.7 % TAE agarose gel. Complete automation NucleoSpin® 96 Blood can be automated on common laboratory workstations. 96 aliquots from the same blood sample have been purified fully automated on a Tecan Genesis™. The average yield is 6.76 ± 0.71 μg with a coefficient of variation (CV) of 10 %. The average purity is 1.97 ± 0.02 (CV: 1 %).
High and consistent yield of genomic DNA Genomic DNA was prepared from 200 μL whole blood using NucleoSpin® 96 Blood on the Tecan Genesis™ fully automated under vacuum. Average yield is 6.76 ± 0.71 μg.
No cross-contamination Due to optimized plate geometry and binding membrane complete processing under vacuum is possible without cross-contaminations. Genomic DNA (48 samples, loaded in a chess-board pattern) was purified from 200 μL whole blood fully automated on a Biomek® 2000. All 96 eluates were used for subsequent PCR (35 cycles). A PCR product is only visible in those wells that contained whole blood in the beginning of the procedure (+). In the other lanes only the primer can be seen.
Removal of PCR inhibitors Genomic DNA from 200 μL blood (96 samples) was isolated on a Tecan Genesis™ 150 and used in LightCycler™ analysis. All samples show a CT value of 23. No inhibition of PCR reactions is observed.
