The Lenti-X Integration Site Analysis Kit contains all the reagents needed for PCR-based identification of lentiviral vector integration sites in transduced cell clones. The first step is the construction of pools of adaptor-ligated genomic DNA fragments, referred to as "libraries". These libraries can then be used for identifying genomic DNA sequences flanking the 5' end of the integrated provirus using long-distance PCR.
Overview
Identify sites of lentiviral integration into the host genome
Simple determination of lentivirus integration sites using GenomeWalker technology
Compatible with most commercially available lentivirus vectors
Complete system including all the necessary enzymes, primers, controls, PCR kit, genomic DNA purification kit, and gel purification kit
Retroviral version also available for detecting integration sites for MMLV and MSCV retroviruses
Identify sites of lentiviral integration into the host genome
Flow chart of the Lenti-X integration site analysis protocol
Flow chart of the Lenti-X integration site analysis protocol. The major steps include construction of three viral integration libraries, PCR amplification of genomic DNA from the libraries, and analysis of PCR products. The gel shows a typical result. Lane 1: HpaI library. Lane 2: DraI library. Lane 3: SspI library. Lane 4: No template control. Lane M: DNA size markers. AP: Adaptor primer. LSP: Lentivirus-specific primer.
