Using the Cellartis Definitive Endoderm Differentiation Kit, cells from the human induced pluripotent stem (hiPS) cell line 18 (ChiPSC18) were differentiated into Definitive Endoderm (DE) cells. These graphs track the relative temporal mRNA expression levels of various markers during differentiation from hiPS cells (starting on day 0) into DE cells (by day 7), as measured by RT-qPCR.

Immunocytochemistry of Cellartis Definitive Endoderm Cells (from ChiPSC18). Cells were thawed, cultured for two days, fixed, and stained for the pluripotency marker OCT4, the definitive endoderm marker SOX17, and the nuclear marker DAPI. Results show that more than 90% of the cell population are definitive endoderm cells and have lost pluripotency.

Cellartis Definitive Endoderm Cells (from ChiPSC18) are human definitive endoderm (DE) cells derived from the human induced pluripotent stem cell line ChiPSC18. The hiPS cells have been differentiated into definitive endoderm cells in vitro to generate a population of >90% SOX17 protein-expressing cells; they are then dissociated into a cell suspension and cryopreserved. These DE cells are suitable for further differentiation into hepatocytes or pancreatic endoderm cells. To differentiate the DE cells into hepatocytes, use the Cellartis Hepatocyte Differentiation Kit (Cat. # Y30050).

During the differentiation process that creates Cellartis Definitive Endoderm Cells (from ChiPSC18), the mRNA expression profile—as measured by RT-qPCR—shows the expected temporal expression of endodermal markers SOX17, FOXA2, FZD5, CXCR4, HHEX, and SOX7; mesoendodermal marker Brachyury; and pluripotency marker OCT4.
