Beta-galactosidase System

Fill in the details below:

The AAVpro Hlper Free System (AAV2-LacZ) provides the plasmids necessary for preparing AAV2 particles to dliever the lacZ reporter gene into mammalian cells. The three included plasmids can be transfected into HEK 293T cells to prepare high-titer virus without a helper virus. This kit includes AAV Extraction Solution for efficient virus isolation without requiring ultracentrifugation.

Overview

  • Produce high-titer AAV particles encoding the lacZ reporter gene under the control of the CMV promoter
  • Enables delivery of the lacZ reporter gene to hard-to-transfect mammalian cells, including proliferating and non-proliferating cells
  • Expression can be detected with the Beta-Galactosidase Staining Kit

Applications

  • Reporter that allows transduced cells to be identified by immunostaining with an anti-beta-galactosidase antibody or by enzyme activity assay
  • Can be used as a positive control

X-gal staining of HT1080 cells infected with AAV2-LacZ

X-gal staining of HT1080 cells infected with AAV2-LacZ

X-gal staining of HT1080 cells infected with AAV2-LacZ. AAV2-LacZ viral particles were prepared using the AAVpro Helper Free System and purified using the AAVpro Purification Kit (AAV2). HT1080 cells were infected with viral particles, and cells were stained using the Beta-Galactosidase Staining Kit.
