HEK 293 Tet-On 3G is a transformed human embryonic kidney-derived cell line that expresses the tetracycline (Tet)-regulated transactivator Tet-On 3G. Inducible expression of any gene can be achieved by transfecting or transducing this cell line with a vector containing your gene of interest under the control of a tetracycline-responsive promoter. Expression is induced by the addition of doxycycline (Dox) to the culture medium. Tet-inducible cell lines are shipped with Tet System Approved FBS.
The Tet-On 3G systems allow inducible gene expression only in the presence of doxycycline (Dox). When Dox binds, the transactivator undergoes a conformational change allowing it to bind tet operator (tetO) repeats within the TRE3G promoter.
Tet-On 3G cell lines were transiently transfected with pTRE3G-Luc, which expresses luciferase under the control of the PTRE3G inducible promoter. The cells were cultured in the presence and absence of Dox, and after 24 hr, luciferase expression was measured. Each cell line demonstrated several thousand-fold difference between induced and un-induced states, even for transient transfection.
